Cooper County is a county located in the state of Missouri. As of 2000, the population is 16,670. Its county seat is Boonville.
According to the U.S. Census Bureau, the county has a total area of 1,477 km² (570 mi²). 1,463 km² (565 mi²) of it is land and 13 km² (5 mi²) of it is water. The total area is 0.91% water.

As of the census of 2000, there are 16,670 people, 5,932 households, and 4,140 families residing in the county. The population density is 11/km² (30/mi²). The racial makeup of the county is 89.05% White, 8.96% Black or African American, 0.36% Native American, 0.23% Asian, 0.02% Pacific Islander, 0.28% from other races, and 1.11% from two or more races. 0.86% of the population are Hispanic or Latino of any race.
